Mechanism of CCL24 Involvement in SSc Pathology.
Following CCL24 binding to CCR3, immune cells recruitment, dermal fibroblast activation, and vascular impairment are seen. Secretion of CCL24 is elevated in the fibrotic tissue resulting in further recruitment of immune cells, increased vascular damage, and additional fibroblasts activation, which, eventually, results in additional secretion of CCL24. Neutralization of CCL24 with CM-101 abrogates its binding to CCR3 and breaks the “vicious cycle” by attenuating inflammation and fibrosis.
